Apparently Quentin Tarantino himself doesn't know; originally it was supposed to be diamonds, but he soon realized that was too mundane. Eventually he just decided to leave it unknown, so each viewer could imagine it for themselves.

It seems that the briefcase might be a reference to a 1955 noir movie called Kiss Me Deadly, which featured a trunk filled with radioactive material.
